农杆菌侵染条件对玉米单倍体遗传转化的影响Effects of Agrobacterium Infection Conditions on Genetic Transformation of Maize Haploid Lines

摘要(Abstract):
以玉米诱导系HHI69诱导玉米自交系获得的单倍体愈伤组织为受体材料,GUS基因为报告基因,采用农杆菌介导法进行遗传转化,对影响转化效率的侵染条件进行了研究。结果表明:继代8 d后的愈伤组织最适宜农杆菌转化;农杆菌菌株EHA105的侵染力好于LBA4404;侵染液浓度OD_(600)为0.7、侵染时间为30 min时,GUS瞬时表达率最高,达到了65.31%。
